HSCT molecular typing

      Human leukocyte antigen(HLA) matching before hematopoietic stem cell transplantation(HSCT) is to determine the HLA compatibility between the donor and recipient. HLA is a cell-surface marker; when cells of different surface markers are mixed together, they would attack each other. HSCT patients generally need SBT matching based on Sanger sequencing techniques and SSO matching through probe hybridization before transplantation.

Sequencing based typing provides HLA typing solutions via direct sequencing method. All of the products utilize the method of BigDye fluorescent dye detection and provide efficient and accurate results. SBT is generally recognized as the “gold standard” for HLA typing. Sequence specific oligonucleotide(SSO), also known as allelic specific oligonucleotide(ASO), through  synthesis of set of oligonucleotide probes complementary to one or more allele sequence(s) and hybridization with PCR amplified products under given conditions, is a technique used to determine the target genotype.

The aim of the HLA match between the donor and recipient is to avoid cell attacks between the cells of the donor and recipient and to avoid post-transplantation rejection reactions and complications such as graft-versus-host disease(GVHD), etc. The higher HLA compatibility between the donor and recipient, the lower the incidence as well as the severity of the rejection and GVHD, and the greater chance of successful transplantation.
